Media Preparation
Aim:
Preparation of L.B Broth and L.B agar to serve as a growth media for bacteria.
Objective:
A nutrient rich media is prepared for the growth of bacteria one is L.B broth and the other
is L.B agar. L.B broth is liquid in nature while L.B agar is solid in nature as agar is being added to
solidify it. The main purpose of the preparation of these two media is to grow bacterial colonies which are
in terms used for study purposes.
Material Used:
• For Luria Broth media
– 2.5g Bacto-tryptone
– 1.25g yeast extract
– 2.5NaCl
– Glass bottle
– Weight balance
– Spatula
• For L.B Agar Plates
– 2.5g Bacto-tryptone
– 1.25g yeast extract

Procedure:
By weighing the required amount of ingredients with the help of measuring balance we
added them into a glass bottle. Than we pour the distilled water in the bottle and made the mixture
volume up to 100ml. After shaking the mixture we sent it for autoclaving.
Autoclaving: The process of getting rid of pre-existing bacteria in the medium.
Observations:
L.B with agar was quite yellow in color while the L.B without agar is somewhat clear.
